An Analysis of Genetic Relationship among Some Chagan-Bamboo Species by Using Fluorescent AFLP

摘要:
利用荧光AFLP技术,选择EcoRⅠ/MseⅠ酶切组合,应用16对( EcoRⅠ+3)/( MseⅠ+3)引物组合进行选择性扩增,检测9个茶竿竹亚属竹种基因组DNA多态性。结果共获得1473条谱带,平均每对引物92条,其中多态性条带1092条,PPB为74.1%。聚类结果表明,正种茶竿竹及其4个变种以相似系数0.78聚在一起,其中铁厘茶竿竹和白水茶竿竹之间的相似系数达到最高为0.88,从分子水平上佐证了铁厘茶竿竹和白水茶竿竹是正种茶竿竹的2个变种。
Abstract:
Fluorescent amplified fragment length polymorphisms ( AFLP ) was used to study the DNA di?versity of 9 Chagan?Bamboo species.Genomic DNA was digested with EcoRⅠ and MseⅠ enzymes and ampli?fied with 16 (EcoRⅠ+3)/(MseⅠ+3) primer combinations.In the 9 samples,16 EcoRⅠ?MseⅠAFLP prim?er combinations produced 1 473 legible bands (92 bands per primer combination),of which 1 092 were poly?morphic bands,the percentage of polymorphic band (PPB) was 74.1%.The cluster analysis of AFLP indicated that Pseudosasa amabilis and its 4 varieties belonged to one small group ( their similar coefficient was 0.78) ,in which the similar coefficient between Pseudosasa amabilis var. ferrea and Pseudosasa amabilis var. peshuiensis was 0.88.This proves that Pseudosasa amabilis var.ferrea and Pseudosasa amabilis var.peshuiensis are two varie?ties of Pseudosasa amabilis.
